Is Google geographic target tool at Google webmaster is problem for region specific domains?

Can it is difficult for region specific domains to rank in google.com?

You cannot set geographic preference for country specific domains. The location is already specified by the domain TLD

No, try to get links from US specific domains.

My problem is a month ago one of my site ranks well in google.uk and google.com but now its disapper form .com. Now to get back in google.com will links from US specific domain works?
