I would like to know if I have one site but in different location(i.e cities and countries) I would like to do the buisness. How would I go for seo strategy for on page and off page point of view.

The best way is to use geo targetted keywords for SEO promotion. You can go for targetted SMO (Social media) like FB fans or twitter followers from any particular region.

One way is Google Adwords....by creating required campaign.
