The first should be seen as a separate domain than the main one... unless it changed not long ago. The folder version will make the blog part of the main domain.

Thanks, so if it's seen as a separate domain and I'm trying to build up a site, that would seem to be a negative? Should I move the blog to the main domain?

You would be better to setup your blog on a sub folder of your main domain, i.e. www.mydomain.com/blog rather than blog.mydomain.com.
Google treats sub domains differently than it does the root domain. If you build up links to blog.mydomain.com then www.mydomain.com would not benefit from link juice being passed in the same way, and vice versa.

@km23: I agree with the sub-folder idea - mainly for the reasons stated by elliott: A sub-domain is essentially a new site; requires linkbuilding and promoting in order to get Google to index beyond the home page, plus the only benefit to your main site is links from the blog (not much use until the blog has backlinks of it's own).
Using a sub-folder means you are adding content/facilities directly to - thus enhancing - your main site (which was - presumably - your original intention) as well as benefitting from the links already flowing into your existing website.
@everyone: If you are going to give your opinion, explain why you recommend that choice... (otherwise you're post is pointless)!

There is not second thought that having blog on sub-folder (.com/blog) is best practice. BUT if your blog is going to handle by your users and if you will allow your users to post content then it is advisable to place it on sub-domain. Because unethical activity like duplicate content, small hidden text won't affect your over all domain as sub-domain is not part of your domain. That's the reason why hub pages turned to sub-domain for all their user generated pages after the Google Panda Update.
